Evaluation of Anti-urolithiatic Potential of Leaves of Alstonia scholaris and its Isolated Pentacyclic Triterpenoids in Ethylene Glycol-induced Renal Calculi Rat Model

Abstract: Introduction: Urolithiasis has afflicted human race since the days of yore. Conventional treatment acts only on associated ailments whereas pharmacologically active phytoconstituents derived from plants are reported to possess lithotriptic as well anti-urolithiatic effect, thus herbal sources need be scientifically explored. Present study was aimed evaluate potential leaves Alstonia scholaris and its isolated pentacyclic triterpenoids: lupeol ursolic acid in ethylene glycolinduced renal calculi rat model. Materials Methods: Leaves were extracted with various solvents, amount triterpenoids each extract quantified using HPTLC. Extract which had higher triterpenoid evaluated for effect. Albino wistar rats either sex divided into 6 groups. All groups except vehicle fed 0.75%v/v glycol 28 days. Treatment standard cystone, low high dose given respective 4th-28th day study. On 28th day, serum creatinine, uric acid, BUN, TNF-α, IL-6 urinary electrolyte levels estimated. Kidneys isolated, one used histopathological other antioxidant assays. Results Discussion: Histopathological examination kidneys revealed destructed morphology due deposition large polymorphic crystals cystic dilatation tubules. Whereas prevented damage crystals. Perturbations nitrogenous wastes, inflammatory mediators electrolytes also prevented. rich significantly arrested oxidative stress inflammation preventing formation calculi.
